Yes, Wing can be used on a machine that has no connection to the internet. It does need to be activated initially, but that can be done through a web browser on another machine. The license dialog at startup provides instructions for this (the manual activation option).
Wing does check for updates by default; this can be disabled via the Auto-check for Product Updates on the User Interface > Advanced page of the Preferences dialog. Also relevant here are Submit Usage Stats and Show Discount Offers, although all of these things should just fail on a non-connected machine and Wing should keep working normally.
